Obesity is characterized by the excessive accumulation of adipose tissue and a body mass index (BMI) above 30 kg/m². Single nucleotide polymorphisms (SNPs) in the ADIPOQ gene have been linked to reduced expression of adiponectin, the protein encoded by this gene, as well as an increased risk of obesity in certain populations. This study investigated the association between the +45 T/G SNP (rs2241766) of the ADIPOQ gene and obesity risk in the Brazilian population. A total of 54 participants were included, classified into 31 obese and 23 non-obese individuals. DNA was extracted and quantified from blood samples, followed by PCR amplification of a 372 bp region containing the +45T/G SNP. Genotyping was performed using the PCR-RFLP method with the restriction enzyme SmaI, and the amplified fragments were analyzed by agarose gel electrophoresis. Statistical analysis was conducted using the chi-square test, with a significance level of p < 0.05. No significant association was found between the +45 T/G SNP and obesity predisposition in the analyzed sample (p = 0.3693). Further studies with a larger sample size will be conducted to enhance the reliability of these findings.
