Polimorfismo do gene ADIPOQ: Análise da sua relação com a obesidade na população brasileira

Resumen

A obesidade é caracterizada pelo acúmulo excessivo de tecido adiposo e por um IMC acima de 30 kg/m². Polimorfismos de nucleotídeo único (SNPs) do gene ADIPOQ têm sido associados com diminuição da expressão da adiponectina, proteína codificada por este gene, e ao risco aumentado de obesidade em algumas populações. Este estudo avaliou a relação do SNP +45T/G (rs2241766) do gene ADIPOQ com o risco de obesidade na população brasileira. Foram analisados 54 participantes, classificados em 31 indivíduos obesos e 23 não obesos. O DNA foi extraído e quantificado de amostras de sangue dos indivíduos, seguido pela amplificação pela PCR de uma sequência de 372 nucleotídeos que contém o SNP +45 T/G. A genotipagem foi realizada pela técnica de PCR-RFLP, utilizando-se a enzima de restrição SmaI. As sequências amplificadas foram analisadas pela eletroforese em gel de agarose. A análise estatística foi realizada pelo teste qui-quadrado, com valor significativo com p≤ 0,05. Não foi detectada associação significativa entre o SNP +45T/G e a predisposição a obesidade na amostra analisada (p = 0,3693). Estudos futuros com um maior número de amostras serão realizados para confirmação dos resultados obtidos.
